In the Deodhar Trophy at Gwalior in March 2005, Sharma made his List A debut for West Zone versus Central Zone. With 24 balls remaining, he batted at number eight and scored 31 not out as West Zone won by three wickets. In the same match, Cheteshwar Pujara and Ravindra Jadeja made their debuts.

Sharma rose to prominence after an unbeaten 142 in 123 balls against North Zone at the Maharanna Bhupal College Ground in Udaipur during the same competition. He travelled to Abu Dhabi and Australia with the India A team before being included to India’s 30-man probables list for the ICC Champions Trophy, however he did not make the final selection. Mumbai went on to win the tournament, with Sharma making a half-century in the final against Bengal in his second innings.

In the Ranji Trophy against Gujarat in December 2009, he got his highest career score of 309 not out. Following the retirement of Ajit Agarkar in October 2013, he was named team captain for the 2013–14 season.

Virat Kohli, India’s captain, was rested for the series against Sri Lanka in December 2017 in order to prepare for India’s visit to South Africa, which began in the first week of January 2018. Sharma was named team captain in his place, and India won the series 2–1, their eighth straight series victory since defeating Zimbabwe in June 2016.

Sharma hit 133 runs in the first match against Australia at the Sydney Cricket Ground on 12 January 2019, but it went in vain as India lost by 34 runs. In one-day internationals, it was his 22nd century. Sharma made 56 runs, including his 8,000th run in one-day internationals, in the fifth and final match in a home series against Australia in Delhi on March 13, 2019. It was his 200th inning of baseball. With 1,490 runs in ODIs, he set a new record for the most runs scored by a batsman in a single year.
