Smith was paralysed as a child due to bone disease, but he recovered his health by the age of ten. He went to Presbyterian Day School in Memphis for grade school and Memphis University School for high school, and as a child, he became an amateur pilot.

Smith enrolled at Yale University in 1962. He wrote a paper for an economics class at Yale that outlined an overnight delivery service. Smith was also a member of the Delta Kappa Epsilon (DKE) fraternity and the Skull and Bones secret society, ultimately becoming its president. In 1966, he earned a bachelor’s degree in economics.

He was a classmate and DKE fraternity brother of future US president George W. Bush during his college years. Smith was also acquainted with future US President George W. Bush. Senator and Secretary of State John Kerry became flying friends and expressed a passion for aviation.

Smith bought a majority stake in Ark Aviation Sales, an aircraft repair business, in 1970, and by 1971, the company had shifted its focus to used jet trade. Smith formed Federal Express on June 18, 1971, with a $4 million inheritance and $91 million in venture capital. The corporation began providing service to 25 cities in 1973, with a fleet of 14 Falcon 20 (DA-20) jets and small packages and papers. His main goal was to create an integrated air-ground system. Smith founded FedEx on the concept of a shipment-based bank clearing house, in which one clearing house was situated in the centre of the representative banks, and all of their delegates were sent to the central location to exchange goods.
